It is located 300 light years from the Bajoran system. (DS9: "Fascination")
This system contained planets capable of supporting life; there was a population on Regulus III. Animal life existed in this system, such as the eel-bird of Regulus V. The Regulan bloodworm also originated here. (TOS: "Amok Time", "The Trouble with Tribbles", DS9: "Fascination")
In 2154, a Vulcan High Command attack fleet massed in this system in preparation for an abortive preemptive strike against the Andorians. The Regulus system was the closest accessible system that was known to be outside the maximum range of Andorian Imperial Guard listening posts. (ENT: "Kir'Shara")

According to the reference work Star Trek: Star Charts, Alpha Leonis (Regulus) is a trinary system. The primary star is a Class B (Blue) star, and the two companion stars are a Class K (Orange) and a Class M (Red). (pg. 56)

The reference work Star Trek: Star Charts presented two contradictory scenarios for the Alpha Leonis system.
Scenario A: Alpha Leonis is a star in the Alpha Quadrant. This star had the alternate name of Deneb. The fourth planet of this system was the homeworld of the Bandi. (pg. 34)
Scenario B: Alpha Leonis is a star in the Beta Quadrant. This star had the alternate name of Regulus. (pg. 56)
Scenario A was most likely a mistake by the author. On page 41, the author identified two systems that were named Deneb. These systems' alternate names were Deneb Kaitos and Alpha Cygni. The appearance of Deneb IV, from page 34, doesn't match the appearance of Deneb Kaitos IV on page 41. Therefore, Deneb IV was a planet in the Alpha Cygni system. It was apparent that the author misidentified the system (Alpha Leonis = Alpha Cygni).

In the episode "Conspiracy", there was a galactic map. This map showed the locations of several stars and planets from Star Trek: The Original Series and Star Trek: The Animated Series. In this map, Regulus was located in a block set between 347 and 348, and five blocks from the galactic edge. (Sol was located in a block set between 348 and 349, and two blocks from the galactic edge.)
In the real world, Regulus is a star in the constellation Leo. This star is located approximately 77.5 light years from Sol.

According to the reference work Star Trek: Star Charts, Alpha Leonis (Regulus) was a destination on a major space lane, and was visited by the Enterprise on her maiden voyage in 2151. (pages 56, 61, 96)
